Applications are invited for a Postdoctoral Scientist to join Dr. Michael Hastings’ and Dr. John O’Neill’s multidisciplinary research groups, in collaboration with Astrazeneca, to investigate the molecular neurobiology of diet-induced neuroinflammation in the hypothalamus, and its modulation by circadian clock mechanisms.
Modern “western” diets are a major cause of poor health. One of the earliest events is the development of neuroinflammatory responses by glial cells in the hypothalamus, leading to mis-regulation of metabolic homeostasis, which further aggravates disease progression. Cell-autonomous and circuit-based circadian clocks modulate inflammatory and metabolic pathways in health and are in turn disrupted by metabolic disease. Loss of circadian homeostasis again aggravates disease progression. However, the links between clocks and metabolism-induced neuroinflammation are poorly understood both at the molecular and neural network levels. We shall explore the consequences of metabolic disorders at the level of the master mammalian clock, the suprachiasmatic nucleus (SCN) alongside the metabolically relevant centres of the mediobasal hypothalamus (MBH). We shall employ spatial transcriptomics to create a hypothalamic cellular atlas, concentrating on transcriptional responses to various pro-inflammatory metabolic loads and their modulation in circadian time. We shall use ex vivo SCN and MBH organotypic cultures to characterise, compare and contrast the inflammatory and circadian consequences of a simulated high-fat diet. Finally, we shall define causal linkages using intersectional genetics and pharmacology.
Candidates should hold a PhD in a relevant biological subject and be experienced in transcriptome bioinformatics and molecular genetics, including RNA extraction, PCR, DNA cloning, genomic sequence analysis, in situ hybridisation etc. Expertise in hypothalamic neurobiology, including working with in vivo mouse models, metabolic or otherwise, is highly desirable.
