A position is available for a Research Assistant for 12 weeks to work on red blood cell morphology and biochemistry in the laboratory of Professor Mark Vickers and Dr Huan Cao in the Institute of Medical Sciences. This project is funded by the University of Aberdeen Development Trust.
The well-known biconcave discoid shape of red blood cells is thought to optimise gaseous exchange across the plasma membrane, but the underlying structural basis is unknown. Using super-resolution microscopy, we have identified that the central ‘hole’ in the centre of the disc is mediated by a molecular bridge between the two underlying membrane skeletons. However, we need further images before this work is ready for publication. The main task of this position is to obtain these images.
Sickle cell disease is an important condition of red blood cells common in areas of the world where malaria is endemic and is caused by a point mutation in the beta-globin gene. We have identified that the presence of sickle cell haemoglobin causes oxidative stress, an important manifestation of which is the production of oxidatively cross-linked protein aggregates marked by high mannose glycans on the cell surface. However, we have not yet identified the glycan-peptide linkages. The second task of this project is to purify these aggregates from blood from patients with sickle cell disease, for further analysis in the chemistry department under the supervision of Prof. Marcel Jaspars.
Salary will be at the appropriate point on the Grade 5 Point 24, £29,619.
